Abstract

Penelitian ini bertujuan untuk mengetahui kadar flavonoid dan aktivitas penghambatan enzim α-amilase terhadap ekstrak dan fraksi daun kersen (Muntingia calabura L.) dengan metode in vitro. Secara empiris, daun kersen (Muntingia calabura L.) dimanfaatkan oleh masyarakat sebagai obat tradisional dalam mengobati beberapa penyakit seperti, sakit kepala, mengatasi peradangan, menjaga kesehatan jantung, mencegah hipertensi, meningkatkan daya tahan tubuh, sebagai antiseptik, mengatasi asam urat, dan sebagai antidiabetes. Hasil kadar flavonoid didapatkan sebesar 20,48 mg QE/g ekstrak daun kersen. Pengujian penghambat enzim α-amilase dilakukan terhadap blangko, kontrol blangko, sampel, kontrol sampel dengan konsentrasi sampel 12,5 ppm, 25 ppm, 50 ppm, 100 ppm, dan 200 ppm. Sampel terdiri dari akarbose, ekstrak etanol, fraksi n-heksan, fraksi etil asetat, dan fraksi air etanol. Pengukuran sampel di lakukan dengan spektofotometri UV-Vis pada gelombang 600nm, didapatkan hasil perhitungan IC50 ekstrak etanol sebesar 123,786 mg/mL, fraksi n-heksan 114,250 mg/mL, fraksi etil asetat 18,138 mg/mL, fraksi air etanol 87,190 mg/mL, dan pembanding akarbose 4,522 mg/mL. Hasil menunjukkan fraksi etil asetat memiliki aktivitas penghambatan yang tertinggi karena senyawa flavonoid yang larut dalam fraksi etil asetat.

This study aims to determine the levels of flavonoids and α-amylase inhibitory activity on extracts and fractions of cherry leaves (Muntingia calabura L.) by in vitro methods. several diseases such as headaches, overcoming inflammation, maintaining heart health, preventing hypertension, increasing endurance, as an antiseptic, overcoming gout, and as antidiabetic. The results obtained flavonoid levels of 20.48 mg QE/g cherry leaf extract. Tests for -amylase enzyme inhibitors were carried out on blanks, control blanks, samples, control samples with sample concentrations of 12.5 ppm, 25 ppm, 50 ppm, 100 ppm, and 200 ppm. The samples consisted of acarbose, ethanol extract, n-hexane fraction, ethyl acetate fraction, and ethanol water fraction. Sample measurements were carried out with UV-Vis spectrophotometry at 600 nm wave, the IC50 calculation results for ethanol extract of 123,786 mg/mL, n-hexane fraction 114.250 mg/mL, ethyl acetate fraction 18.138 mg/mL, ethanol water fraction 87,190 mg/mL, and a comparison of acarbose 4.522 mg/mL. The results showed that the ethyl acetate fraction had the highest inhibitory activity because the flavonoid compounds were soluble in the ethyl acetate fraction..
