N-HEXAN EXTRACT AND FRACTION INVOLVEMENT TEST,
ETHLACETATE FRACTION, KERSEN LEAF WATER FRACTION

(Muntingia calabura L) AGAINST BACTERIA
Staphylococcus aureus ATCC 25923

Cherry leaves (Muntingia calabura L.) contain many flavonoids, namely
flavones, flavanones, and flavans so that they can be antibacterial. This study aims
to determine the inhibitory ability of extracts and fractions of N-Hexane, Ethyl
Acetate fraction, Kersen Leaf Water fraction (Muntingia calabura L) against
Staphylococcus aureus ATCC 25923 and to determine the most active fraction to
inhibit bacteria.
Extract and fraction tests were carried out using the diffusion and dilution
method on the extract sample macerated with 96% ethanol and then continued
with the fractionation process. Tests using the diffusion method using a
concentration of 1%,5%,10%,15%,20% using a positive control ciprofloxacin
0.5% and the dilution method with concentrations of 50%, 25%, 12.5%, 6.25%,
3.12% , 1.56%, 0.78%, 0.39%, 0.195% and 0.098%.

The results of the antibacterial activity test showed that the extract, N-
Hexane, Ethyl Acetate, and Water fractions had inhibitory power against bacteria.

The most active fraction in this study was the ethyl acetate fraction with the
inhibitory power of the ethyl acetate fraction at a concentration of 20% with an
average inhibition zone of 11.6 mm. The results of the dilution test of the Ethyl
Acetate Fraction with KBM at a concentration of 12.5%.
