Abstract
Latar Belakang: Resistensi antibiotik golongan B-laktam disebabkan perubahan penicillin-binding proteins (PBP). PBP1 Staphylococcus aureus merupakan protein transpeptidase biosintesis dinding sel. Beberapa penelitian ekstrak daun kirinyuh (Chromolaena odorata L) menunjukkan aktivitas penghambatan terhadap staphylococcus aureus secara in vitro. Tujuan: penelitian ini memprediksi potensi penghambatan senyawa bioaktif daun kirinyuh terhadap protein PBP1 staphylococcus aureus. Metode: penelitian metode in silico dengan bantuan aplikasi VegaZZ, PyRx, Discovery Studio, SwissADME dan ToxTree. Hasil: senyawa 7-angeloylretronecine, 9-angeloylretroene, rinderine, intermedine, rinderine N-oxide, intermedine N-oxide, 4,5,6,7-tetramethoxyflavone, odoratin, isosakuranetin, quercetin, luteolin, apigenin, acacetin, sinencetin, quercetagetin, chlorogenic acid, p-coumaric acid, p-hydroxybenzoic acid, protocatechuic acid, dan vanillic acid mampu berikatan dengan protein PBP1 dengan rentang nilai AG −5,3 hingga −8,1 kcal/mol dan nilai RMSD 0,543–2,157 A melibatkan residu SER A:314, SER A:349, dan ASN A:370, yang sama dengan residu interaksi ligan natif oksasilin. Senyawa 7-angeloylretronecine, 9-angeloylretroene, p-coumaric acid dan vanillic acid memenuhi parameter Lipinskis Rule of Five dan farmakokinetik. Senyawa p-hydroxybenzoic acid, protocatechuic acid, p-coumaric acid dan vanillic acid memenuhi paremeter cramer rules. Kesimpulan: senyawa p-coumaric acid berpotensi sebagai inhibitor protein PBP1 pada staphylococcus aureus.
