Demam tipoid merupakan penyakit infeksi serius serta merupakan penyakit endemis yang sampai saat ini masih menjadi masalah dan disebabkan oleh bakteri Salmonella typhi. Penggunaan obat antibiotik untuk pengobatan penyakit karena infeksi bakteri dapat menimbulkan resistensi. Salah satu tumbuhan yang dapat dimanfaatkan sebagai obat tradisional dan memiliki khasiat sebagai antibakteri yaitu tumbuhan pepaya (Carica papaya L). Kandungan alkaloid karpain dalam biji pepaya diduga memiliki efek sebagai antibakteri. Penelitian ini dilakukan untuk mengetahui potensi aktivitas antibakteri fraksi n-Heksan, fraksi etil asetat dan fraksi air biji pepaya (Carica papaya L) terhadap bakteri Salmonella typhi ATCC 13311. Penelitian ini bersifat eksperimental. Proses ekstraksi dan fraksinasi menggunakan metode cair-cair. Pengujian aktivitas antibakteri fraksi n-Heksan, fraksi etil asetat dan fraksi air biji pepaya (Papaya California L) dilakukan pada konsentrasi 10%, 25%, 50%, dan 75% dengan tiga kali replikasi. Metode pengujian menggunakan difusi cakram dan dilusi. Kontrol positif menggunakan Ciproflaxacin 5 µg dan kontrol negatif menggunakan pelarut DMSO 1%. Analisis data diameter daya hambat dilakukan menggunakan uji One Way ANOVA. Fraksi teraktif pada pengujian difusi adalah fraksi etil asetat dengan konsentrasi 75% dan daya hambat sebesar 12,23 mm. Nilai KHM dan KBM pada pengujian dilusi yaitu pada konsentrasi 18,75%. Hasil uji One Way ANOVA data terdistribusi normal dibuktikan dengan nilai (P>0,05). Fraksi biji pepaya (Papaya California L) memiliki aktivitas antibakteri terhadap bakteri Salmonella typhi ATCC 13311.

Typhoid fever is a serious infectious disease and is an endemic disease which is still a problem and is caused by the bacterium Salmonella typhi. The use of antibiotic drugs for the treatment of diseases due to bacterial infections can cause various problems. One of the plants that can be used as traditional medicine and has antibacterial properties is papaya (Carica papaya L). The content of karpain alkaloids in papaya seeds is a compound that is thought to have an recistence. This study was conducted to determine the potential antibacterial activity of the n-Hexane fraction, ethyl acetate fraction and water fraction of papaya seeds (Carica papaya L) against Salmonella typhi ATCC 13311 bacteria. This research is experimental. The extraction and fractionation process uses the liquid-liquid method. The antibacterial activity of the n-Hexane fraction, ethyl acetate fraction and water fraction of papaya seeds (Carica papaya L) were carried out at concentrations of 10%, 25%, 50%, and 75% with three replications. The test method uses disc diffusion and dilution. positive control using Ciproflaxacin 5 µg and negative control using 1% DMSO solvent. Inhibition diameter data analysis was carried out using the One Way ANOVA test. The most active fraction in the diffusion test was the ethyl acetate fraction with a concentration of 75% and an inhibitory power of 12.23 mm. The MIC and MBC values in the dilution test were at a concentration of 18.75%. The results of the One Way ANOVA test data are normally distributed as evidenced by the value (P> 0.05). The papaya seed fraction (Papaya California L) has antibacterial activity against Salmonella typhi ATCC 13311 bacteria.
