ANTIBACTERIAL ACTIVITY TEST OF THE EXTRACT AND N-HEXAN
FRACTION, ETHYL ACETATE FRACTION, WATER FRACTION
FROM PEGAGAN LEAVES (Centella asiatica (L.) Urb) AGAINST THE
BACTERIA Escherichia Coli ATCC 25922 BY DIFFUSION AND
DILUTION

Pegagan leaves (Centella asiatica (L.) Urb) is one of the plants that can be
used as an antibacterial. Pegagan leaves contain chemical flavonoids, tannins and
saponins. The purpose of this study was to determine the antibacterial activity of
ethanol extract, n-hexane fraction, ethyl acetate fraction and water fraction from
pegagan leaves and to determine Minimum Inhibitory Concentration (MIC) and
Minimum Kill Concentration (MBC) of the most active fraction of pegagan leaf on
the growth of Escherichia coli ATCC 25922.
Pegagan leaf powder was macerated using 96% ethanol, then fractionated
using n-hexane, ethyl acetate, and water as solvents. The extract, n-hexane fraction,
ethyl acetate fraction and water fraction were tested for antibacterial activity using
diffusion and dilution methods. Diffusion method with concentration 1%, 5%, 10%,
15%, 20% and dilution method with concentration 50%, 25%, 12.5%, 6.25%,
3.12%, 1.56%, 0, 78%, 0.39%, 0.195% and 0.098% against Escherichia coli ATCC
25922.
The results of the antibacterial activity test using the diffusion method
showed that the extract, n-hexane fraction, ethyl acetate fraction and water fraction
of pegagan leaf could inhibit bacterial growth in the presence of inhibitory power.
The 20% ethyl acetate fraction was the most effective because it had the highest
average inhibition of 13.67 mm. The results of the dilution method test showed that
the MBC value of the ethyl acetate fraction was 12.5%. Based on the results of the
study, it can be concluded that the ethyl acetate fraction is the most active fraction.
